Dissertation in short:

Refractive errors (REs) arise when the focal length of optics show a mismatch with the axial length of the eye. Myopia is the most common type of RE and has shown a dramatic increase in prevalence over recent decades. Given current trends, 50% of the world’s population is expected to be myopic by 2050 and 10% will suffer from high myopia. The rising global prevalence of myopia is accompanied by a serious health burden, as myopia is associated with an increased risk for serious eye diseases such as glaucoma, myopic macular degeneration, cataract, and retinal detachment 3,4. Almost 500 RE-associated loci and annotated candidate genes have been identified in the last decade but functional evidence remains elusive. In this thesis we aim to provide the first functional evidence supporting genetic associations found in GWAS studies for RE.
