Below is a brief summary of the dissertation:
Unlocking Cell Secrets in Real Time
Cells are constantly moving, dividing, and changing, but studying rare behaviors under a microscope has long been like finding a needle in a haystack. This thesis introduces real-time image analysis tools designed for a high-throughput microscope developed in our lab: the ultrawide field-of-view optical (UFO) microscope.
By combining fast image processing with precise laser phototagging, our system automatically catches rare cellular events while cells are still alive. We can track fast-moving breast cancer cells, spot abnormal divisions linked to chromosomal instability as they happen, and isolate those specific cells for genetic sequencing. We also built deep learning models that combine brightfield and fluorescence images to predict how a cell will divide before it actually does.
Together, these methods link live microscopic video directly with genetic analysis, opening up new ways to study cancer progression and dynamic cell behaviors as they unfold.
