Brief summary of the doctoral thesis:
UBE3A is an E3 ubiquitin ligase and is known to play a critical role in brain development. A testament to its importance during neurodevelopment is that the absence of functional UBE3A leads to a severe neurodevelopmental syndrome, called Angelman Syndrome (AS). Similar to several other proteins in our cells, different UBE3A protein isoforms are expressed, however it is not clear whether and how each UBE3A isoform contributes to the protein’s functions in health and disease. The aim of this thesis was to investigate the isoforms of UBE3A and their properties – expression, subcellular localization and function – and to understand their individual roles in brain development and by extend in AS. To achieve this goal, we adopted a multifaceted approach, intertwining research in mouse models to study brain development with molecular and biochemical methods.
