Pompe disease is a slowly progressive muscle disease which can be treated with enzyme replacement therapy (ERT). However, the effect of ERT to skeletal muscles seems larger than the effect to respiratory muscles and there is a large variation in long-term treatment effects between individual patients.

In this thesis we used MRI to detect small changes in the motion and shape of the diaphragm that were not detected using regular pulmonary function tests. These MRI markers can help to detect early diaphragmatic weakness and to evaluate diaphragmatic weakness over time. Careful monitoring of individual patients is necessary to support treatment decisions, especially when hopefully stem cell therapies will be available.
