Brief summary on the aim of the doctoral thesis:
Diabetic retinopathy (DR) is a common complication of type 2 diabetes mellitus (T2DM). Chronic hyperglycemia is the main initiating factor in the development of DR. This leads to endothelial damage, abnormal vascular permeability, reduced blood supply and local hypoxia, initiating the formation of new blood vessels in the retina. Inflammation plays a crucial role in both early and late stages of DR, contributing to damage and impaired healing of retinal vessels. Myelomonocytic cells including macrophages and monocyte-derived pro-angiogenic cells (PAC) are involved in initiating endothelial activation and inflammation, as well as in the pathological angiogenesis observed in disease situation. Hyperglycemia drives pro-inflammatory activation of these cells, further contributing to chronic inflammation in endothelial cells. The aim of this thesis is to scrutinize aspects of diabetes- and DR-related conditions, in particular involving the lipoproteins Lp(a) and LDL, and their influence on retinal endothelial cells (REC) and monocyte-related PAC.
