Below is a brief summary of the dissertation:
This thesis is focused on new therapeutic options for pediatric acute lymphoblastic leukemia (ALL) and chronic myeloid leukemia (CML). We provide the results of two ‘first-in-child’ clinical trials, developed in the context of Pediatric Investigational Plans (PIPs), with the final aim to inform the Regulatory Authority and obtain approval of inotuzumab ozogamicin (InO) and bosutinib for pediatric use.
The majority of drugs we use in pediatric oncology are not approved for the indication, and sometimes not even for use in children. In fact, development and approval of new agents for pediatric cancers lags behind adult development. New regulations in Europe and U.S. are enacted to ensure commitment of companies to develop drugs in children, but current incentives are still insufficiently attractive for pharmaceutical companies. Clinical trials in children, especially early phase 1-2 studies, are challenging due to the small number of patients with cancer and relapsed or refractory disease.
